Is it possible to have a PI under two different institute’s?
We have a PI who’s institute is SKCC Translational Research, but the Distribution Protocol receiving institute is Thomas Jefferson University, Distribution Site is SKCC Translational Research.

No, it is not possible to assign a user to two institute. In your case, SKCC seems like a site within TJU. So you can maybe consider creating multiple sites and assigning users to multiple sites are allowed in OpenSpecimen. If you are still facing issues, contact support for further help.
